Generation and Maturation of Human Monocyte-derived DCs

[Abstract]  Dendritic cells (DC) are antigen-presenting cells, which play a critical role in the regulation of the adaptive immune response. They act as a bridge between the innate and the adaptive immune systems. An approach to study their function and potentiality is to generate DC-like cells by culturing CD14+ monocyte-enriched peripheral blood mononuclear cells (PBMC). In the presence of GM-CSF and IL-4, these cultures give rise to large numbers of DC-like cells. Generating human-DC from PBMC is a useful tool to study biological functions of human DC. Rabbit IgG Conjugation to Dynabeads

[Abstract]  This method couples rabbit IgG (or any other proteins serve as affinity reagent) to the surface of magnetic beads (Brand name: Dynabeads). The amine and thiol groups of amino acid residues on the protein are covalently linked with the epoxy group on Dynabeads. The coupled IgG beads can be stored at 4°C for 6-12 month with no obvious loss in reactivity. [摘要]  该方法是将兔子的IgG（或者可作为亲和试剂的任何蛋白质）与磁性凝胶珠的表面结合（商品名：免疫磁珠），蛋白质上的胺类和硫醇类氨基酸基团与免疫磁珠的环氧基共价键结合，结合的IgG可以在4°C的条件下保存6-12个月，而不会引起明显的活性丧失。
